One of the three closely-related members of the TGF-β family. Human TGF-β1 cDNA encodes a 390 aa precursor with a 29 aa signal peptide. A furin-like convertase cleaves the precursor to form a 249 aa latency-associated peptide (LAP) and a C-terminal 112 aa mature TGF-β1. Disulfide-linked mature TGF-beta1 is dissociated from the LAP upon activation.
StemRD, produces active TGF-β1 in human 293 cells in Xeno-free medium. After a series of chromatography, the protein is purified to near homogeneity. Activity is determined by using a SMAD reporter gene assay with EC50 0.1 – 1.0 ng/mL.
